Abstract.  1. The fungus Neotyphodium lolii forms a symbiotic relationship with its grass host Lolium perenne (perennial ryegrass). The fungus benefits from access to plant nutrients and photosynthate, whereas the plant benefits from acquired chemical defence against herbivory.
2. This study examined the potential for endophyte-mediated plant defences to influence interactions between fall armyworm Spodoptera frugiperda , and the entomopathogenic nematode Steinernema carpocapsae and clarified biological mechanisms underlying the observations made.
3. In laboratory and greenhouse experiments, S. frugiperda larvae were fed endophytic or non-endophytic L. perenne then exposed to S. carpocapsae or injected with the nematodes' symbiotic bacteria Xenorhabdus nematophila .
4. In all instances, S. frugiperda larvae fed endophyte-infected grass suffered significantly lower mortality than those fed non-endophytic plants. Although larvae fed endophyte-infected grass often had significantly lower biomass than those fed uninfected grass, these differences did not account for altered susceptibility to S. carpocapsae .

Endophyte-mediated reductions in herbivore susceptibility to the nematode pathogen represent a herbivore adaptation that effectively turns the tables on both plant and natural enemy by reducing the virulence of the nematodes' symbiotic bacteria while expanding the temporal window of herbivory.  相似文献

Abstract Pigment decomposition, oxygen evolution and CO2 fixation were measured in the cyanobacterium Phormidium uncinatum after infection with cyanophage LPP-1, under light and dark conditions. A gradual decrease in para benzoquinone supported O2 evolution, chlorophyll a and phycocyanin level were noticed after 6 h of infection. These results demonstrated decreased photosynthetic activity of the host P. uncinatum prior to the start of LPP-1 multiplication. Metabolic inhibitor investigations confirmed that the cyanophage LPP-1 multiplication was independent of host photosynthesis.  相似文献

Endophytic fungi belonging to the genus Neotyphodium often form symbiotic associations with grasses. The host plants usually benefit from the association with an endophyte. Presence of the symbiont may increase host resistance to infection by some pathogens. However, the exact mechanism of the lower susceptibility of endophyte‐infected plants to diseases is still unclear. Growth chamber trials were conducted to determine whether (a) tall fescue plants infected with the endophyte Neotyphodium coenophialum (E+) are more resistant to sheath and leaf spot disease caused by Rhizoctonia zeae than endophyte‐free (E?) plants, and (b) R. zeae growth inhibition is associated with endophyte presence. Tall fescue genotypes, each symbiotic with a genetically different native endophyte strain, were inoculated with isolates of R. zeae. The tillers infection by R. zeae, density of endophyte hyphae and content of total phenolic compounds in tillers were studied. Antifungal activity of the N. coenophialum towards R. zeae, Rhizoctonia solani, Bipolaris sorokiniana and Curvularia lunata was also investigated in dual‐culture assays. For Tf3, Tf4, TfA2 and TfA9 tall fescue genotypes, the E+ plants had reduced R. zeae infection. In the Tf9 and Tf8085 genotypes, R. zeae infection was similar for both E+ and E? plants. The strongest effect was observed for the Tf4 endophyte. A strongly positive correlation (r = 0.94) occurred between endophyte hyphal density and disease index across all tall fescue genotypes. Dual‐culture assays showed no inhibitory interaction between the seven endophyte strains and the R. zeae isolates; however, some endophytes inhibited R. solani, B. sorokiniana and C. lunata. Endophyte presence increased the production of phenolic compounds by the host grasses. The level of phenolics also differed significantly depending on the time of analysis after inoculation of plants by R. zeae. The results indicate that N. coenophialum can suppress disease severity caused by R. zeae infection. The mechanism of higher resistance of E+ plants is likely not based on direct inhibition such as antibiosis or competition. Thus, the induction of specific mechanisms in the host plant, for example, production of phenolic compounds, seems to be the main way of providing resistance to the grass by the endophyte.  相似文献

Tall fescue (Festuca arundinacea Schreb.) plants infected by the fungal endophyte Neotyphodium coenophialum (Morgan-Jones & Gams) (Glenn et al., 1996) often perform better than noninfected plants, especially in marginal resource environments. There is a lack of information about endophyte related effects on the rhizosphere of grasses. In a greenhouse experiment, four endophyte-infected (E+) tall fescue clones (DN2, DN4, DN7, DN11) and their endophyte-free (E–) forms were grown in limed (pH 6.3) Porter soil (low fertility, acidic, high aluminum and low phosphorus content, coarse-loamy mixed mesic Umbric Dystrochrept) at three soil P levels (17, 50, and 96 mg P kg-1 soil) for five months. Excluding the genotype effect, endophyte infection significantly increased cumulative herbage DM yield by 8% at 17 mg P kg-1 soil but reduced cumulative herbage DM yield by 12% at 96 mg P kg-1 soil. With increased P availability in the soil, shoot and root DM, and root/shoot ratio in E+ plants were significantly less when compared to E– plants. Endophyte infection increased specific root length at 17 and 50 mg P kg-1soil. At soil P level of 17 mg P kg-1soil, E+ plants had significantly higher P concentrations both in roots and shoots. Similar relationships were found for Mg and Ca. E+ plants had significantly higher Zn, Fe, and Al concentration in roots, and lower Mn and Al concentration in shoots when compared to E– plants. Ergot alkaloid concentration and content in shoot of E+ plants increased with increasing P availability in the soil from 17 to 50 mg P kg-1 but declined again at 96 mg P kg-1 soil. Ergot alkaloid accumulation in roots increased linearly with P availability in the soil. Results suggest that endophyte infection affects uptake of phosphorus and other mineral nutrients and may benefit tall fescue grown on P-deficient soils. Phosphorus seems also to be involved in ergot alkaloid accumulation in endophyte-infected tall fescue.  相似文献

Initial studies of grass–endophyte mutualisms using Schedonorus arundinaceus cultivar Kentucky‐31 infected with the vertically transmitted endophyte Epichloë coenophiala found strong, positive endophyte effects on host‐grass invasion success. However, more recent work using different cultivars of S. arundinaceus has cast doubt on the ubiquity of this effect, at least as it pertains to S. arundinaceus–E. coenophiala. We investigated the generality of previous work on vertically transmitted Epichloë‐associated grass invasiveness by studying a pair of very closely related species: S. pratensis and E. uncinata. Seven cultivars of S. pratensis and two cultivars of S. arundinaceus that were developed with high‐ or low‐endophyte infection rate were broadcast seeded into 2 × 2‐m plots in a tilled, old‐field grassland community in a completely randomized block design. Schedonorus abundance, endophyte infection rate, and co‐occurring vegetation were sampled 3, 4, 5, and 6 years after establishment, and the aboveground invertebrate community was sampled in S. pratensis plots 3 and 4 years after establishment. Endophyte infection did not enable the host grass to achieve high abundance in the plant community. Contrary to expectations, high‐endophyte S. pratensis increased plant richness relative to low‐endophyte cultivars. However, as expected, high‐endophyte S. pratensis marginally decreased invertebrate taxon richness. Endophyte effects on vegetation and invertebrate community composition were inconsistent among cultivars and were weaker than temporal effects. The effect of the grass–Epichloë symbiosis on diversity is not generalizable, but rather specific to species, cultivar, infection, and potentially site. Examining grass–endophyte systems using multiple cultivars and species replicated among sites will be important to determine the range of conditions in which endophyte associations benefit host grass performance and have subsequent effects on co‐occurring biotic communities.  相似文献

Many grasses in the subfamily Pooideae develop symbioses with Neotyphodium fungal endophytes, which exist widely in nature. The stably symbiotic relationship not only ensures accessible nutrients required by Neotyphodium fungal endophytes, but also significantly increases the resistance of host grasses to biological stresses through the production of secondary metabolites. Previous studies show that infected grasses with endophytic fungi have prominently enhanced resistance to pests, plant diseases, companion plants and other biological stresses. Grass endophytic fungi show remarkable resistant to at least 79 species of pests from three classes; arachnida, nematode and insecta, and to at least 22 species of pathogenic fungi. Although the biotechnological application of endophytic fungi in grass breeding for variety selection and quality improvement has progressed well, opportunities remain for further exploring the use of fungal endophytes among different host grasses coupled with the examination of genetic stability of Neotyphodium in novel host grasses. In the future application of endophytic fungi as a bio-control method, researchers should not only consider specificities of host grasses, but also need to have comprehensive analysis and knowledge about the mutual relationships among grasses, endophytic fungi and ecological environments, which will help use endophytic fungi to better serve humanity.  相似文献

Background and Aims Neotyphodium lolii is a fungal endophyteof perennial ryegrass (Lolium perenne), improving grass fitnessthrough production of bioactive alkaloids. Neotyphodium speciescan also affect growth and physiology of their host grasses(family Poaceae, sub-family Pooideae), but little is known aboutthe mechanisms. This study examined the effect of N. lolii onnet photosynthesis (Pn) and growth rates in ryegrass genotypesdiffering in endophyte concentration in all leaf tissues. • Methods Plants from two ryegrass genotypes, Nui D andNui UIV, infected with N. lolii (E+) differing approx. 2-foldin endophyte concentration or uninfected clones thereof (E–)were grown in a controlled environment. For each genotype xendophyte treatment, plant growth rates were assessed as tilleringand leaf extension rates, and the light response of Pn, darkrespiration and transpiration measured in leaves of young (30–45d old) and old (>90 d old) plants with a single-chamber openinfrared gas-exchange system. • Key Results Neotyphodium lolii affected CO2-limited ratesof Pn, which were approx. 17 % lower in E+ than E– plants(P < 0·05) in the young plants. Apparent photon yieldand dark respiration were unaffected by the endophyte (P >0·05). Neotyphodium lolii also decreased transpiration(P < 0·05), but only in complete darkness. There wereno endophyte effects on Pn in the old plants (P > 0·05).E+ plants grew faster immediately after replanting (P < 0·05),but had approx. 10 % lower growth rates during mid-log growth(P < 0·05) than E– plants, but there was noeffect on final plant biomass (P > 0·05). The endophyteeffects on Pn and growth tended to be more pronounced in NuiUIV, despite having a lower endophyte concentration than NuiD. • Conclusions Neotyphodium lolii affects CO2 fixation,but not light interception and photochemistry of Pn. The impactof N. lolii on plant growth and photosynthesis is independentof endophyte concentration in the plant, suggesting that theendophyte mycelium is not simply an energy drain to the plant.However, the endophyte effects on Pn and plant growth are stronglydependent on the plant growth phase.  相似文献

Poor livestock performance on tall fescue is linked to infection by a fungal endophyte that enhances grass resistance to stress, including herbivory, while producing ergot alkaloids toxic to vertebrate grazers. Novel ‘safe’ endophyte/grass associations produce no ergot alkaloids yet retain stand persistence, but they could be more susceptible to insect outbreaks. We tested the hypothesis that grass‐feeding insects are more abundant in novel endophyte pastures compared with those containing common endophyte. Above‐ and below‐ground herbivores were sampled across two growing seasons in pastures containing common strain endophyte (KY31), novel endophytes (MaxQ or AR584) or endophyte free. We also sampled natural enemies as an indicator of possible tritrophic effects. With a few exceptions, numbers of predatory, chewing (grasshoppers, crickets, caterpillars) or sucking insects (leafhoppers, planthoppers) were similar in common and novel endophyte pastures. Contrary to our hypothesis, Aphrodes spp. leafhoppers were more abundant in KY31 than other pasture types in 2008; their nymphs also were more abundant in KY31 than in MaxQ in 2009. Adults (but not total numbers or nymphs) of another leafhopper, Psammotettix striatus, were less abundant in KY31 than other pasture types in 2009. Popillia japonica and Cyclocephala spp. grubs did not differ in density, weight or instar among the endophyte‐containing associations. In feeding assays, armyworm (Mythimna unipuncta) survival was higher on MaxQ and NE9301, and weight gain was higher on NE9301, suggesting armyworm outbreaks could be more severe on such grasses, but that result may not be attributable solely to alkaloids because common strain and endophyte‐free grasses did not differ in either parameter. Caterpillar abundance did not differ among pasture types in the field. Our results suggest that re‐seeding common strain endophytic pastures with livestock‐safe novel endophyte/grass associations to alleviate fescue toxicosis is unlikely to promote markedly higher populations of plant‐feeding insects.  相似文献

The combinations of three genotypes of Lolium perenne with and without (i) infection by the fungal endophyte Neotyphodium lolii, (ii) infection by ryegrass mosaic virus and (iii) one of five different forms of abiotic stress were studied in pot experiments in a glasshouse. The five abiotic stress treatments were (i) low pH (compared with ‘optimal’ pH), (ii) cutting plants to a height of 1 cm (compared with 5 cm), (iii) shading (compared with no shading), (iv) cutting plants at 2‐weekly intervals (compared with 6 wk) and (v) low nitrogen applied (compared with ‘high’ nitrogen applied). On average, over the five experiments, the accumulated herbage dry weight was 10% more for N. lolii‐infected plants than uninfected, 22% more for virus‐free plants than infected, and 265% more for ‘unstressed’ plants than for plants with abiotic stress. The effects of N. lolii infection on plant growth when the plants were under abiotic or biotic stress were not consistent.  相似文献

Clavicipitaceous fungi of the genus Neotyphodium occur widely as mutualistic, systemic, seed‐borne infections in festucoid grasses. Grass infection by these fungi is associated with the presence of a range of secondary metabolites (SM), several of which have been demonstrated to confer to the plant resistance against herbivorous vertebrates and insects. An initial experiment demonstrated that endophytic infection by Neotyphodium can influence the utilisation of grasses by Deroceras, with feeding preferences and impact on plant yields affected differentially by endophytes with different SM profiles. The role of Neotyphodium SM in feeding preferences of Deroceras slugs were then evaluated in artificial diets. Among the indole diterpenoids tested, lolitrem B was demonstrated to reduce feeding, while diets containing paxilline, lolitriol, α‐paxitriol and β‐paxitriol tended to be preferred over that of untreated diet. The pyrrolopyrazine alkaloid peramine had no effect. Among the ergopeptine alkaloids tested in the diets, ergotamine and ergovaline were demonstrated to be phagostimulatory. These results with artificial diets were generally consistent with Deroceras reticulatum preferences among plants of known Neotyphodium endophyte strain and SM profile. Deroceras slugs obtained from sites containing contrasting frequencies of Neotyphodium‐infected grasses, exhibited differential responses to Neotyphodium SM incorporated into artificial diet. This study demonstrates that infection of grasses by different isolates of Neotyphodium endophytes differentially influence herbivory by molluscs, reflecting their SM profile. These results offer an explanation for variable acceptability of grasses to molluscs and their importance in the diet of molluscs in the field reported in previous studies in both natural and agricultural systems. Neotyphodium endophytes potentially offer novel approaches to management of mollusc pests in agricultural gramineous crops.  相似文献
